MOLECULAR DETECTION OF STAPHYLOCOCCUS PSEUDINTERMEDIUS ISOLATED FROM OTITIS IN DOGS

The aim of this study was to isolation and identification of Staphylococcus Pseudintermedius by 16S ribosomal gene sequence. Out of ninety samples were collected from dogs infected with otitis from different areas in Baghdad province in a period from (November2021 to February 2022).These samples were collected under hygienic condition and transferred immediately in to 5ml of fresh tryptic soy broth and transported to the college laboratory, these swabs were cultured on different media (selective and differential) such as (Staphylococcus 110 and mannitol salt agar) and were incubated at 37 C° for 24 hours. These isolates were recognized depending on their morphological shape ,size ,color ,depending on grams stain and subjected to different biochemical tests, and confirmed by using VITEK2 system the basement of this study was to selection the most dominant Staphylococcus Spp. and to be more confirmed by detection of ribosomal rRNA by using PCR, the Staphylococcus isolates were 35(38.9%) furthermore these isolates were more confirmed by Vitek 2 system and the results were 10 (28.5%) isolates of Staphylococcus Pseudintermedius, 7(20%) isolate of S.sciuri ,5 (14.3%) S.warnerei 5(14.3%) S.xylosus,4(11.5%) S.haemolyticus 3(8.5%) isolates of S.aureus and 1(2.8%) isolates of S.hominis, the most dominant Staphylococcal spp .was Staphylococcus Pseudintermedius which was selected to be more confirmed by detection of 16S ribosomal rRNA genes sequence using PCR, which were deposited in to the national Center Biotechnology information (NCBI) Gene Bank, and was tested for antimicrobial sensitivity test using 11 antibiotics discs , and the results was all isolates 10 (100%) resistant for pencilliin, and 5(50%) of isolates were resist to methicillin, azithromycin, vancomycin,4(40%) were resist to oxacillin, chloramphenicol, clindamycin, 2(20%)to cefixime, 6(60%) to tetracycline ,3(30%) to netilmicin and 2(20%) to amoxicillin\ clavunalic acid. Conclusion: Staphylococcus pseudintermedius is the most frequent species isolated from otitis externa in dogs and was successfully identified by 16 sRNA gene sequence using PCR, the antimicrobial pattern appeared multi drug resistant isolates that indicate difficult of treatment of dogs.
